Abstract/Contents: | "A total of five different organisms across three genera associated with take all root rot on ultradwarf bermudagrass greens were evaluated for pathogenicity and in vitro fungicide sensitivity assays. Gaeumannomyces graminis (Gg), Gaeumannomyces sp. (Gx), Gaeumannomyces graminicola (Ggram), Candidacolonium cynodontis (Cc), and Magnaportheopsis cynodontis (Mc) were individually and co-infested on sterile oats and used to inoculate 'Champion' bermudagrass cores (4.4 cm diameter) in a greenhouse. Basil necrosis, turfgrass density, root length, root weight, and clippings weight were evaluated. Take all root rot organisms were evaluated using in vitro fungicide sensitivity assays against 10 fungicides from 3 different chemical classes. Fungicide concentration were 0, 0.01, 0.1, 1.0, and 10 ppm. Experimental units were incubated in constant darkness at 25°C and two-way colony diameter was measured on day 3 (Gg and Ggram) and day 6 (Cc and Mc). The effective concentration resulting in a 50% reduction of growth compared to the control (EC50) and relative mycelial growth (RMG) were determined using the probit procedure. 'Champion' bermudagrass inoculated with Ggram had the highest basil necrosis (33.3%) and lowest plant density. Basil necrosis and plant density were negatively correlated with an r2 of 0.80. Gg was the only other organism that showed an increase in basil necrosis compared to the control. However, when Cc and Mt were co-inoculated there was an increase in basil necrosis and decrease in plant density. Take all root rot organisms differed in fungicide sensitivity. All isolates evaluated against succinate dehydrogenase inhibiting fungicides consistently had the highest EC50 and RMG values. The lowest EC50 and RMG values were observed when isolates were grown on azoxystrobin amended media. Differences in pathogenicity and fungicide sensitivity within and across chemical classes provide novel insight into biological factors of take all root rot organisms." |
