Abstract/Contents:"Dollar spot caused by the fungus Clarireedia spp. is the most destructive and economically important fungal disease of turfgrass species, especially on golf course fairways and putting greens. It impacts the turf quality by diminishing the aesthetic value and playability. There are ~16,000 golf courses in the U.S., and 54% of golf course managers execute calendar-based fungicide applications during the growing season, which costs approximately $15,000 annually per golf course. There is a need to establish a phenotyping system for rapid and accurate quantification of visible symptoms as well as early invisible indicators of disease activity, that can lead to more timely and effective fungicide treatments. For that, a detailed investigation of the Agrostis-Clarireedia spp. interaction is necessary. Here, we infected 2 creeping bentgrass (Agrostis stolonifera) cultivars, Crenshaw (highly susceptible to dollar spot), Providence (susceptible), and 1 colonial bentgrass (Agrostis capillaris) cultivar, BCD (tolerant), with 2 Clarireedia spp. isolates, Clarireedia jacksonii, and Clarireedia monteithiana, to evaluate dollar spot disease development, from inoculation, penetration, infection, incubation, reproduction, to propagation. Up to 2 weeks after inoculation, we performed daily analyses of microscopic observation, quantification of absolute abundance of Clarireedia spp., primary and secondary metabolite profiling, ROS measurement (H2O2 and superoxide) to understand the temporal progression of the Agrostis-Clarireedia spp. interaction. These results will support the development of a field level high-throughput imaging system, that can assist in reducing fungicide costs as well as negative impacts on the environment."
