Abstract/Contents: | "This research focuses on a novel approach for ectotrophic root-infecting (ERI) fungal detection associated with bermudagrass decline (BD). Two summer core aerifications (July 2017 and July 2018) were used to collect root material from two golf greens at Mississippi State University Golf Institute. Greens were selected by BD symptomology based on superintendents suggestion of his healthiest (Green 2) and most symptomatic (Green 12) greens. Root material was collected from multiple areas of interest (AOI) within Green 2 (n=68) and Green 12 (n=66). Fungi of interest included the causal agent of BD, Gaeumannomyces graminis, as well as undescribed Gaeumannomyces sp., novel Candidacolonium cynodontis, and Magnaporthiopsis cynodontis. Two multiplex qPCR assays were used to identify the selected fungi, one determining presence and quantity of the two Gaeumannomyces spp. and ultradwarf bermudagrass (UDB) root, and the other determining presence and quantity of C. cynodontis, M. cynodontis, and UDB root. Multiple complexes of ERI fungi were identified in AOIs in both greens. Gaeumannomyces graminis, G. sp., C. cynodontis, and M. cynodontis were identified in 28, 78, 50, and 90% of AOIs, respectively, within Green 2 in 2017 as well as 0, 97, 51, and 100% in 2018. While inoculum densities for G. graminis, G. sp., and C. cynodontis were similar across years; inoculum density of M. cynodontis increased from 2017 to 2018 in Green 2. In Green 12, G. graminis, G. sp., C. cynodontis, and M. cynodontis were identified in 95, 20, 27, and 98% of AOIs, respectively, in 2017; as well as 48, 70, 51, and 97% in 2018. The inoculum density of G. graminis decreased from 2017 to 2018 in Green 12, whereas G. sp. and M. cynodontis increased across years and inoculum density of C. cynodontis was unchanged. Overall, colonization of roots by these select fungi occurred regardless of plant health." |
