Abstract/Contents:"Genetic transformation offers possibilities to make great improvement in zoysiagrass (Zoysia japonica Steud.) performance. Therefore, an orthogonal experimental design with genotypes, growth regulators, and Cu2+ concentration was carried out for quick screening of the best culture medium formula for callus induction and embryogenic callus production. The results indicated that there are significant genotypic effects on callus induction and embryogenic callus production. Among four varieties tested, 'Zenith' performed the best. The highest embryogenic callus production was obtained with 5 mg L-1 of 2,4-D, 0.1 mg L-1 of Kin or BA. Cu2+ concentration at 2.5 mg L-1 yielded the highest callus induction rate, but 5 mg L-1 showed to be toxic for callus growth. A test for particle bombardment of embryogenic calli was also successful and 9 individual transgenic plants were obtained after hygromycin selection and GUS confirmation assay."
