Abstract/Contents:"Selection of superior turfgrass genotypes in vitro is complicated by the unpredictable impact of the environment on plant performance. Although microculturw selection systems confer greater environmental control, callus tissues often fail to mimic whole plants for selected traits. A continuous, uniform, whole plant microculture maintenance system for Kentucky bluegrass (Poa pratensis L.) was established to facilitate realistic selection. The system was applied to an in vitro disease assay for root and crown rot diseases, and to selection of salt and drought tolerant plants. Co-culture of whole plant microcultures and virulent fungal isolates permitted clear observation of root lesion development over time, without destroying samples. Virulence of the fungal pathogens and freedom from competing microorganisms were guaranteed by using inoculum isolated from infected, aseptic whole plant cultures. Drought or salt stress conditions were induced in culture by adjusting the composition of the growth medium, and osmotic adjustment in grass leaf cells was measured to assess tolerance. Stress symptoms (lesion development, chlorosis, and stunting) were objectively rated using an adeptation of video image analysis."
