Abstract/Contents:"The causal agent of a severe patch disease occurring on Tifdwarf hybrid couch (Cynodon dactylon × C. transvaalensis) golf greens at Ocean Shores Country Club on the north coast of New South Wales, Australia, was identified as Gaeumannomyces graminis var. avenae, the most common cause of take-all patch in turfgrasses. Glasshouse testing showed that it was pathogenic to Tifdwarf hybrid couch but not to couch (Cynodon dactylon) cultivars such as Wintergreen, Greenlees Park and Common. Molecular diagnosis using a test based on the polymerase chain reaction (PCR) with specific primers confirmed the identity established by morphological characteristics such as simple (unlobed) hyphopodia and ascospore length [86 (110) 138 μm]. The DNA method was quicker (1 week vs 8 weeks) and more reliable than the production of perithecia in infected plant tissues or agar and has potential for the development of a rapid diagnostic test for take-all patch disease in turfgrasses."
