Abstract/Contents:"Waitea circinata var. circinata (Wcc), is the causal agent of brown ring patch of annual bluegrass (Poa annua) that was recently discovered in the western U.S. Isolates from CA, CT, IL, MA, NV, NY, OR, RI and WA were identified as Wcc based on colony morphology and a previously published RFLP method using HapII to differentiate Waitea spp. Some of the Wcc isolates idetified by morphology produced the characteristic two bands at around 475 and 170 bp whereas others produced the same bands but also had apparently uncut genomic DNA. Sequencing the ITS rDNA confirmed that some isolates where heterozygous at the HapII restriction site which explained our results. [moreover], the sequencing reactions failed in about half of the reads. This was due to two indels at poly T positions found after cloning and sequencing 3 to 5 clones. Intra-species variability in the ITS was low (~2%) but phylogenetic analysis clearly separates Wcc from W. circinata var. oryzae and W. circinata var. zeae. The results of this work indicate that Wcc is wide spread across the U.S. as a pathogen of annual bluegrass and not just found in the western U.S. as previously thought. ITS variability also indicates that potential problems in identification may occur using RFLP."
