Abstract/Contents: | "Four clones of white ash (Fraxinus, americana L.) selected for rapid microshoot proliferation were used. Proliferating axillary shoots (20 mm long) were rooted by pulsing in MS medium with 20% sucrose, 0.7% Difco Bacto agar, 5μuM NAA, and 1μuM IBA for 8 days in darkness, then transferring to microplant medium containing MS with 1/4-strength macro-salts (1/4 MS), 10% sucrose, and 0.7% agar. When roots were visible (6-10 days after transfer to microplant medium), microplants were transferred to microplant medium containing tall fescue (Festuca elatior L.) extracts. Cut leaves from tall fescue in 21.6 liter pots in a greenhouse were prepared by grinding or soaking 10g (f.w.) and bringing volume in 100 ml liquid 1/4 MS. Soil leachates were also tested. Extracts were filtered, then serially diluted with microplant medium. After 4 weeks in vitro, primary and secondary root length were reduced by extracts from 5 and 10 g ground leaves per 100 ml medium. However, extracts from soaked, cut leaves stimulated root growth by 30% when applied at a rate of 5g per 100 ml." |
