Abstract/Contents:"Two soluble enzymes have been isolated and partially purified from tall fescue that possess major invertase activity. We distinguish these two enzymes by their relative transferase activities. While "Invertase S" only begins to synthesize the frutosylsucrose triaccharide at 0.5 M sucrose, "Invertase Q" sythesizes significant amounts at 0.1 M sucrose. This is of particular interset to us as we characterize fructan biosynthesis in this species. To date, the major transferase activity of Invertase Q does not resolve from the hydrolytic activity by ion exchange chromatography, Chromatofocusing, lectin affinity chromatography, size exclusion chromatography, hydroxyapatite chromatography, or hydrophobic interaction chromatography. These enzymes are being described with reference to pH optima, temperature optima, kinetic constants, and substrate specificity. No vacuolar metabolites or ions have been found to significantly effect Invertase Q activity. We are currently investigating the kinetics of the triaccharide sythesis."
