Abstract/Contents:"The objective of this study was to define the use of Evan's blue as a vital stain for root hairs of perennial grasses. Three warm-season grasses, (Cynodon dactylon [L.]Pers. x C. transvaalensis Davy) 'Tifway' bermudagrass, (Paspalum notatum Flugg.) 'Pensacola' bahiagrass, and (Stenotaphrum secundatum [Walt.] Kuntze) 'Texas Common' St. Augustinegrass, were grown from sprigs in medium-course quartz sand in Dee pots (6.25 cm diam., 25 cm deep) for 100 d under grasshouse conditions. Plants were watered twice daily and fertilized via weekly applications of a Hoagland's nutrient solution. Evan's blue, as a root hair vital stain gave a statistically confirmed 100%, 94.8%, and 95.8%, correlation to 2M sucrose induced cellular collapse for tifway, Pensacola and Texas Common, respectively. When using this stain to determine whether or not roots could be stored and their root hair viabilities determined, roots severed at the crown were placed in 0.05 phosphate buffer, pH 7.0, and stored at 4 C with no light sheilding. Unfortunately, algal and fungal growth, plus root rot and nematode activity nullified accurate data collection. In a second storage study, roots severed at the crown were placed in sterile 0.05 N phosphate buffer, pH 7.0, and stored in the dark at 4 C. There was a significant increase in both root hair viability and number between 0 and 3 weeks of storage for all grasses. There was no significant difference between 3 and 6 weeks of storage for all grasses."
