| Abstract/Contents: | 1989 study to :1) develop an agar-cellulose medium for study of Japanese beetle (JB) and southern Masked chafer (SMC) grub feeding ecology and to facilitate oral delivery of possible feed deterrents to white grub and 2) test for feeding deterrency of loline and ergot alkaloids to 3rd instar JB grubs using agar-cellulose medium. The first objective was achieved by comparing various concentrations of several sugars in a medium containing 4% agar and 4% cellulose. Addition of amino acids or ascorbic acid at various concentrations did not significantly increase feeding of JB grubs; however, the addition of 0.01 M or 0.025 M ascorbic acid to 0.1 M sucrose in the medium did significantly stimulate feeding of SMC grubs relative to sucrose alone. Addition of N-formyl- & N-acetyl loline, in concentrations comparable to those that occur in fescue roots (50-100 ug/g), to agar/cellulose medium containing 0.1 M sucrose significantly deterred feeding by 3rd instar JB grubs. Ergot alkaloids also deterred feeding of 3rd instar JB grubs when incorporated into the same agar medium, but at concentrations higher than naturally found in roots. Peramine was not a significant feeding deterrent. |
