| Abstract/Contents: | "Strains of Psuedomonas syringae resistant to copper (Cu), streptomycin (Sm), or both were isolated from cottonwood, ornamental pear, and willow exhibiting tip dieback and cankers. Resistant strains contained a single plasmid which ranged in size from 60-200 kilobase pairs (kb) among strains. Plasmids encoding both Cur and Smr were transferred by conjugation from their respective donors to a sensitive strain of P. syringae at frequencies ranging from 1.4 by 10 -2 [to the -2 power] to 9.3 by 10 -8 [to the -8 power]. Strong homology at high stringency was demonstrated between the P. syringae plasmids and the Smr genes from the broad-host-range plasmid RSF1010. This homology was used to clone the P. syringae Smr gene(s) on a 3.8 kb PstI fragment in pBluescript II SK." |
