Abstract/Contents:"Juvenile cyst nematodes (Heterodera sp. and Punctodera sp.) were discovered in turf samples from golf course greens in the western United States. In the western USA, cyst nematodes are not common in golf course greens, but their numbers could increase due to the absence of efficient nematicides. In the past, cyst nematodes in golf course greens in the United States have been identified based on morphology, especially that of the mature cyst. When cysts are not recovered, identification must be based on the infective juveniles (J2). Due to morphological variation among J2 within a species, identification with molecular tools is necessary. Juvenile cyst nematodes were extracted from soil using a mist extractor and individual J2s were lysed. PCR was conducted with primers specific for the ITS region of nematodes. The resulting PCR products were sequenced. We have identified two species, Heterodera ustinovi and Punctodera punctate. This is the first record of H. ustinovi in golf course greens in the western USA."
