| Abstract/Contents: | "Kentucky bluegrass (Poa pratensis L.) seed produced in northern Minnesota is often contaminated with quackgrass seed. Currently, there are no selective herbicides which control quackgrass in Kentucky bluegrass seed production fields. Our objectives were 1) to establish a tissue culture system for Kentucky bluegrass and 2) to select for tolerance to sethoxydim or fluazifop in tissue culture. Immature inflorescences were aseptically excised and plated onto MS2 media. Callus was initiated from 26 varieties of Kentucky bluegrass. Fast-growing, friable, embryogenic callus was identified and plants were regenerated. Embryogenic callus was exposed to increasingly inhibitory concentrations of sethoxydim and fluazifop. Plants regenerated from callus that survived on 8 uM sethoxydim and 3.2 uM fluazifop media showed no increased tolerance to the herbicides at half the recommended field rates." |
