Abstract/Contents:"Though turfgrass species and cultivars within species have been identified by conventional methods, a reliable method is needed for accurate identification. In order to identify species and cultivars within turfgrass species, restriction fragment length polymorphisms (RFLP) analysis of chloroplast and nuclear DNA was conducted. Restriction fragment patterns of chloroplast DNA (cpDNA) were separated by agarose gel electrophoresis. RFLP analysis of nuclear DNA was carried out using the wheat ribosomal RNA gene (rDNA) as a probe. RFLP of cpDNA separated turfgrass species, whereas RFLP of nuclear rDNA separated cultivars within species. RFLP of cpDNA and nuclear rDNA may be used to identify species and cultivars within species of turfgrasses. Dendrogram constructed based on RFLP of cpDNA and nuclear rDNA enabled us to estimate phylogenetic relationship among turfgrasses, suggesting that phylogenetic relationship among turfgrasses can be studied by RFLP analysis of cpDNA and nuclear rDNA. According to these results, tall fescue was found to be more closely related to Lolium species than to the fine-leaved fescue. It suggested that classification of tall fescue should be reconsidered."
