| Abstract/Contents: | "A new method of DNA fingerprinting has been developed and used for legal proof of genetic individualism between turfgrass species and cultivars. This method, DNA Amplification Fingerprinting (DAF) creates bar graph-type amplified fragment length polymorphisms (AFLPs) when amplifying DNA with short oligonucleotide primers of arbitrary sequence. This method constitutes the state-of-the-art technology for protecting breeders rights, identifying genes of economic importance, and for pathogenic and taxonomic identification, among other uses. The procedure is fast, needs only very small amounts of DNA, is low cost compared to RFLP-based fingerprinting, and uses no radioactivity. Genome analysis by DAF was shown to produce specific amplification patterns which could be analyzed visually and densitometrically after PAGE separation and silver staining. DNA amplification of turfgrass DNA using single or multiple primers was used to enhance understanding of genome divergence, cultivar identity, and genetic mapping of relevant adaptive gene loci." |
