Abstract/Contents:"Establishment of the efficienct protoplast culture system is the prerequisite for the practical application of the transformation and cell fusion techniques for the improvement of Japanese lawngrass (Zoysia japonica Steud). This study was conducted to determine basic conditions for the isolation, culture of protoplasts and plant regeneration from cultured protoplasts of Japanese lawngrass. Suspension culture was established from embryogenic callus with the capacity to regenerate plants. Embryogenic callus was derived from sterile mature seeds of commercial Japanese lawngrass on Linsmaier and Skoog (LS) medium wtih 2,4-dichrolophenoxyacetic acid (2,4-D) at 5 mg L-1. Embryogenic callus was selected visually under microscope and proliferated in liquid N6 medium with amino acids (N6-AA medium) to give fine suspension culture suitable for the isolation of protoplasts. Protoplasts were isolated in an enzyme mixture with pectolyase Y23 and successfully divided when cultured in K8P medium using 2 mg L-1 of 2,4-D at the density of 106 ml-1. Plants were regenerated by transferring the callus derived from protoplasts to Murashige and Skoog (MS) medium without plant growth regulator and by incubating at 28 C under light (40 uEm-2s-1, 12h photoperiod) for two months. Ten plantlets were planted to the soil and successfully grown to maturity."
