Abstract/Contents:"In recent years variety discrimination and identification have been achieved in a range of agricultural crops by means of electrophoretic techniques. Previous work on Lolium cultivars has shown the potential of these methods for variety analysis, but the time required for protein separation, typically five to six hours, was found to be a major disadvantage. In this research, the potential of a rapid electrophoretic technique applied to the genus Lolium was investigated. 65 seed lots of six Italian ryegrass cultivars (Lolium multiflorum Lam.), 18 perennial ryegrass (L. perenne L.) and four hybrid ryegrass varieties (L. x boucheanum Kunth) were examined. Protein extraction in a buffer containing Tris-HCl (pH 6.8), sodium dodecylsulphate, dithiothreitol and glycerol, was accelerated by sample heating, and denatured proteins were separated in a horizontal discontinuous buffer system on 8-18% SDS-polyacrylamide-porosity gels. Due to electrical conditions and electrophoretic apparatus chosen, an advantageous shorter electrophoretic run of 75 min. was possible. It was therefore possible to carry out a complete analysis within five hours. Differences in the resulting seed protein banding patterns enable differentiation of the three Lolium species and all cultivars analysed. Identification of 17 out of 20 unknown samples after electrophoretic analysis showed the potential of this technique for variety identification purposes. The application of identical electrophoretic methodology, however, was essential to get reliable and reproducible results."
