Abstract/Contents:"Twenty-seven zoysiagrass (Zoysia spp.) lines were screened for fluazifop-P-butyl tolerance under greenhouse conditions. The herbicide rate that caused 50% biomass reduction (GR50) and the rate that caused 50% injury (IR50) were calculated for each line. The three most tolerant and the three most susceptible lines were selected and then used for studying the physiological mechanisms responsible for fluazifop-P-butyl tolerance. Organophosphate insecticide phorate was used as a cytochrome P450 inhibitor to test the involvement of cytochrome mediated metabolism in fluazifop-P-butyl tolerance, but no changes in injury were observed when comparing phorate treated and non-treated plants after fluazifop-P-butyl application. Sequencing of the ACCase gene confirmed none of the seven previously reported mutations conferring resistance to ACCase-inhibiting herbicides in other species was present in any of the six zoysiagrass lines studied. Absorption and translocation rates were studied using C14 radiolabeled fluazifop-P-butyl. No difference in absorption was detected among lines at each individual harvest timing (1, 3, 7 and 10 days after treatment, DAT). However, radiolabeled fluazifop-P-butyl translocation to root and shoot tissue was higher for one of the susceptible lines, compared with that of other five lines. Additionally, this line exhibited the shortest time to reach 50% absorption (3.1 DAT). In metabolism experiments using treated leaves, at 7 days after application, two metabolites in addition to the parent fluazifop-P-butyl and fluazifop acid were detected in one of tolerant zoysiagrass lines. Our results suggest that the diversity in tolerance to fluazifop-P-butyl in zoysiagrass germplasm is most likely the result of a combination of different non-target site mechanisms such as uptake, translocation and compartmentation after absorption as well as detoxification through metabolism or conjugation."
