Abstract/Contents:"Many grass species are known to host fungal endophytes in a mutualistic relationship. We have found that the endophyte Acremonium typhinum produces an unusual endoprotease during symbiotic infection of the grass Poa ampla. This protease is highly active in the presence of SDS. It is a thiol-containing serine protease and is localized to a crude membrane fraction. We have investigated three other endophyte-infected Poa spp. and found that each endophyte also produced a similar protease. Expression of this protease may be important in endophytic infection of Poa spp. since similar activity has not been detected in endophyte-infected Loliuum perenne (perennial ryegrass) or Festuca arundinacea (tall fescue). Using Sephyacryl 300 column chromatography, we have obtained a partially purified preparation of the protease. On SDS polyacrylamide gel electrophoresis the protease migrates at an extremely high apparent molecular weight of >300,000."
