USGA Summary Points: | The fungal endophyte (Epichloë festucae) that infects strong creeping red fescue produces an abundant antifungal protein that is not found in most Epichloë species. It may be involved in the disease resistance observed in endophyte-infected strong creeping red fescue. We have expressed and purified modified forms of the antifungal protein in the bacterium E. coli The modified forms of the antifungal protein had activity against the model test fungus Neurospora crassa. We successfully carried out a gene knockout of the E. festucae antifungal protein gene using the CRISPR-Cas9 gene editing technology. Isolates with the knockout will be reintroduced into strong creeping red fescue to evaluate susceptibility to dollar spot disease. |
