Abstract/Contents: | "Poa annua, or annual bluegrass, is a prolific grass species considered a weed in most turfgrass situations. Along with its proliferative characteristics, reports of herbicide resistant P. annua are steadily increasing. P. annua has been found resistant to ten different herbicide modes of action and some cases of multiple resistances within the same population. In an effort to quantify the spread of herbicide resistant P. annua across the country, the USDA Specialty Crops Research Initiative (SCRI) provided funding to 16 professors in the ResistPoa Project. This project has focused on screening populations suspected of herbicide resistance with eleven different herbicides, and those that show a considerable level of resistance were selected and sequenced for known target site mutations found in EPSPS, ALS, and psbA genes. Populations were sequenced using either capillary or amplicon sequencing, depending on the complexity of the gene, and then analyzed to determine if target site resistance was present. If populations did not contain target site resistance, said populations were forwarded to the next stage of the experiment determine if non-target site resistance was the resulting cause of herbicide resistance. With the populations suspected resistant, 77.98% of ALS, 57.53% of EPSPS, 76.86% of psbA, and 47.9% of populations with multiple resistances were sequenced. From the samples sequenced, 65.65% of ALS, 73.81% of EPSPS, 37.63% of psbA, and 34.29% of populations with multiple resistances contained a target site mutation." |
