Abstract/Contents:"Conditions for maximizing transient expression and the activity of four promoter/intron constructs in protoplasts of Japanese lawngrass (Zoysia japonica Steud.) were investigated. It was found from the transient expression of the GUS reporter gene that polyethylene glycol (PEG)-mediated plasmid uptake largely depended on PEG concentration, plasmid quantity, and heat shock treatment of protoplasts before the addition of PEG. In the transient assay with four constructs, rice actin 1 promoter showed the highest level of transgene expression, suggesting that it is efficient for use in Japanese lawngrass transformation."
