Abstract/Contents:Cell wall preparations from Italian ryegrass endosperm cells grown in tissue culture were analysed for residual glucans and other polysaccharides. Stationary phase cells contained 3.0% protein, 14% 1,3-glucose and 49% 1,4-glucose residues: xylose, arabinose and galactose were also present. The amino acid composition of the protein is given. The cell wall preparations were fractionated by successive extraction with water at 40 deg C, 8 M urea, 0.5 M KOH, 4.27 M KOH and 6 M NaOH/0.81 M H3BO3. The 3 main polysaccharides identified in the fractions were 1,3:1,4- beta -glucan, arabinoxylan and/or galacto-arabinoxylan and cellulose. The mixed-linked glucans and arabinoxylans were present in the 8 M urea and 0.5 and 4.27 M KOH fractions. The presence of hydroxylamine-sensitive linkages in the wall was confirmed.
