| Abstract/Contents: | "Phosphinothricin N-acetyl transferase gene (pat) was cotransformed into zoysia (Zoysia japonica Steud.) protoplasts with a hygromycin-resitance marker gene by polyethylene glycol (PEG)-mediated direct gene transfer. Selection with a high concentration (400 mg / l) of hygromycin yielded a number of resistant calli and about one hundred plants were generated from these resistant calli. The polymerase chain reaction and Southern hybridisation analysis revealed that all of tested plants contained one or more introduced hygromycin-resistance gene and more than half of them also contained one or more introduced pat gene. Application of herbicide to the plants of pat-transformed zoysia showed no necrosis, confirming the expression and functional activity of the part gene in transgenic plants." |
