Abstract/Contents:"Herbicide resistant transgenic creeping bent lines were evaluated under field conditions in 1994 and 1995. These lines were obtained from biolistic and protoplast transformations with chimeric bar genes encoding phosphinothricin acetyltransferase. Vegetatively multiplied plants were planted in randomised plots. Herbicide treatments were at 0, 1x (0.84 kg ha⁻¹) and 3x (2.52 kg ha⁻¹) field rates of Glufosinate and replicated three times. All the transgenic plants that displayed complete herbicide resistance in greenhouse tests demonstrated a high level of herbicide resistance in field tests. They were unaffected by both 1x and 3x rates and looked green and healthy. Treated control plants were killed within 5-10 days after herbicide application. 37 plants, from biolistic transformation and 33 plants from protoplast transformation showed a high level of herbicide resistance. Seeds were harvested from vernalised transgenic plants pollinated with seed-grown control plants in a containment greenhouse. Transgenic seedlings were able to germinate in the presence of herbicide while control seeds of the same cultivar did not germinate in Petri dish germination tests. After transplanting to soil, transgenic seedlings were completely resistant to a 2 mg/ml Herbiace (equivalent to 3.36kg ha⁻¹ Bialaphos) treatment in a greenhouse test. Our results show that herbicide resistance was inherited in the seedlings."
