Abstract/Contents:"Random Amplified polymorphic DNA (RAPD) analysis of nuclear DNA was conducted to identify turfgrass cultivars. Fourteen cultivars of Kentucky bluegrass (Poa pratensis L.) and sixteen cultivars within four species of bent (Agrostis spp. L.) were studied. These cultivars were characterised by RAPD markers amplified using fifty kinds of primers including 8 - 10 mer of synthesised oligonucleotides and commercial primers (10 mer), purchased from Operon Inc.. Some RAPD markers were shown to be specific to some cultivars. We cloned and sequenced such a specific DNA band from the cultivar 'Touchdown' of Kentucky bluegrass. DNA amplification with the primers, which were constructed based upon the sequence information of the specific DNA band, were found in only 'Touchdown' among fourteen cultivars of Kentucky bluegrass. Our results showed that the cultivar 'Touchdown' could be identified by PCR amplification using a specific primer set. Therefore, RAPD analysis and the PCR detection system (AMFLP) can be utilised for the identification of cultivars of Kentucky bluegrass and also would be applicable to cultivars of other turfgrass species."
