Abstract/Contents:"Self-incompatibility in Cynodon dactylon (L.) Pers. is well documented, but little is known of its cytological basis. Accordingly, research was undertaken to characterise the self-incompatibility response within the species relative to pollen germination and rate and extent of pollen tube growth. Controlled cross- and self-pollinations were effected in the greenhouse using five genotypes previously characterised for self-fertility (3.6 to 8.7%) and cross-fertility (39 to 91%). Pollen germination and pollen-tube growth were examined using pistils dissected from florets collected 2.5, 5, 7.5, 10, and 24 h post-pollination. Pistils were stained with 0.1% aniline blue and examined using fluorscence microscopy. Pollen germination exceeded 80% within 2.5 hours of pollination regardless of genotype or pollination mode. Pollen tubes readily penetrated stigmatic tissues, but growth rate upon entry was contingent on genotype and pollination mode. Only a few pollen tubes of 'SS-3', 'SS-13', '70-3' and '72-6' reached the micropyle within 24 h post self-pollination. Pollen tubes of 'Zebra' reached the micropyle within 2.5 h post self-pollination. Pollen tube growth in all genotypes was observed in the micropylar region within 2.5 h post cross-pollination, indicating no cross-incompatibilities. The results suggest a gametophytic system whereby the pollen tube growth response is determined by pollen genotype."
