Abstract/Contents:"We have constructed a RFLP linkage map of Zoysia spp. from 105 self-pollinated progenies derived from a putative interspecific hybrid between Zoysia japonica and Z. matrella, cv. Miyako, with the aim to identify QTLs related to the winter leaf-colour. In the field trials of the self-pollinated progenies, colour scores (0 : dead ~ 5 : dark green) in winter varied significantly. A RFLP analysis of the self-pollinated progenies with 41 genomic clones (ZG), 20 cDNA clones with mRNAs extracted from leaves (ZCL), and 21 cDNA clones from rhizomes (ZCS) of Z. japonica identified 95 RFLP loci in the self-pollinated progenies. The linkage map comprised 18 linkage groups with 84 loci. The other eleven loci could not be associated to a particular linkage group. In total, the linkage map covered 1220cM of zoysia grass genome. The RFLP linkage map was used to establish QTLs related to the winter leaf-colour in the self-pollinated progenies using a single-point ANOVA at each locus. Four QTLs located in the linkage group VI, VII, and XII, respectively, were detected at P< 0.05. The means of winter leaf-colour in matrella genotypes at QTLs in the linkage group VII and ZG160 were higher than those of japonica genotypes, whereas QTLs derived from Z. japonica in the linkage group VI and XII were increased winter leaf-colours."
