| Abstract/Contents: | Enzymatic (takadiastase) and acid methods for the determination of total non-structural carbohydrates (TNC) were compared using rhizomes of Panicum repens and Cynodon dactylon and commercial starch. The action of different conc. of acid (0.2-3.0 N H2SO4) on fructose, inulin, cellulose and glucose were also studied. With the grasses only 0.2 N H2SO4 gave similar results to takadiastase, while 1 or 1.5 N H2SO4 gave much higher values. It was concluded that the acid method can be inaccurate especially in plants rich in easily hydrolysable structural carbohydrates and that if such a method must be used it should be preceded by fractionation of the plant material. |
