| Abstract/Contents: | "In vivo assays for nitrate reductase activity (NRA) have been used extensively in studies of plant N metabolism, whereas in vivo assays for nitrite reductase activity (NiRA) have not often been employed. The objective of this study was to optimize assay conditions for in vivo NRA and NiRA in Kentucky bluegrass (Poa pratensis L.) leaves. Maximum NRA was obtained by using a medium containing 100 mM potassium phosphate buffer (pH 7.5), 390 mM 2-propanol and 50 mM potassium nitrate. Lower pH and higher concentrations of propanol inhibited NRA. Dark and anaerobic conditions were essential for assays of NRA. Under illumination, nitrite was absorbed by leaf tissues and reduced by nitrite reductase to ammonium, which accumulated in the presence of methionine sulfoximine. Based on this observation, an in vivo NiRA assay was developed. The use of these methods for studying turfgrass N metabolism will be discussed." |
