| Abstract/Contents: | "The seedling root fluorescence (SRF) test has been used for seventy years to distinguish annual ryegrass (Lolium multiflorum Lam.) from perennial ryegrass (L. perenne L.). The SRF test does serve as a general indicator of annual ryegrass contamination of perennial ryegrass seed lots. The SRF character, however, is a poor indicator of genetic contamination between these two interfertile species. DNA testing would provide a fast, and accurate alternative to the SRF test. Screening of over 700 RAPD primers in a backcross population using Bulked Segregant Analysis has yielded no useful linkages with the SRF character. Esterase isozyme analysis indicates that the reported esterase difference between annual and perennial ryegrass may be bue to tissue specific expression of the esterase locus. A comparative genetics approach using markers linked to genes influencing vernalization requirement in barley may provide useful information in the development of a replacement test for SRF." |
