| Abstract/Contents: | "Creeping bentgrass (Agrostis palustris Huds., cv. Penncross) was genetically transformed by biolistic insertion of the mannitol 1-phosphate dehydrogenase [mtlD] gene. The mtlD gene is reported to aid in drought and salinity tolerance via osmoregulation through the production of mannitol. Mannitol accumulation in tissues of regenerated clones and non-transformed Penncross was evaluated under salt and drought stress conditions. Salt stress was induced for 4 days by irrigation with 100 mM NaCl, followed by irrigation with tap water for 3 days. Drought stress was induced by subjecting a separate set of plants to 10% soil moisture before a 3 day recovery period. Carbohydrates were extracted three times by heating whole plants to 70 C in 80% ethanol for 30 minutes. Extracts were dried then rehydrated using millipore water. Mannitol content was measured using high-performance liquid chromatography (HLPC) using a Shodex sugar SC1101 column." |
