Abstract/Contents: | Changes in sucrose and fructosan hydrolysing activity (SHA and FHA) associated with the insoluble fraction in the haplocorm of timothy cv. Climax were examined during regrowth after cutting. SHA increased during the 1st 2 days after cutting, decreased until the 4th day and then remained almost constant at about 1.5 times the initial level. FHA was not affected by cutting. The opt. pH was 4.5 for SHA and 6.0 for FHA. SHA was greatest at around 30 deg C. All the saccharides tested were hydrolysed except inulin, suggesting that beta -fructofuranosidase, alpha -glucosidase, beta -glucosidase and cellulase were bound to the insoluble fraction. Sucrose was most actively hydrolysed. Sucrose hydrolysing enzymes were partially released from the insoluble fraction by treatment with papain, but not by 1M NaCl, 0.25 M borate buffer, cellulase, bromelain and pronase. SHA remaining in the insoluble fraction was increased only by treatment with cellulase. |
