Abstract/Contents:"Plants that 'acclimate' are able to survive exposure to otherwise lethal low temperatures. One major change occurring during cold acclimation is the increase in the membrane-lipid tri-unsaturated fatty acid linolenic acid. In 'Midiron' and "Quickstand', two relatively cold tolerant bermudagrass cultivars, this increase occurs sooner and to a greater degree than in U3 and Arizona Common, two relatively cold-sensitive genotypes. This study focuses on the structure and organization of the genes encoding enzymes involved in fatty acid biosynthesis, namely, stearoyl-ACP desaturase, linoleoyl desaturase and linolenyl desaturase. cDNA clones of three desaturase genes were obtained by RT-PCR. Genome organization was studied by Southern hybridization analysis, and showed that the desaturases are encoded by small gene families. Restriction fragment length polymorphisms were observed between the cultivars, and in some cases there was a correlation between the banding pattern and a cultivar's phenotype. Expression of the desaturase genes in different tissues and time points during cold acclimation was studied by Northern blotting."
