Abstract/Contents: | "Bermuda grass etched-line Marafivirus (BELV) infects Bermuda grass and certain other gramineous plants in Morocco and Iran. It causes reduced growth and etching of leaves in Bermuda grass and a fine mosaic in others. The Iranian isolate was purified by differential and density-gradient centrifugation. Viral RNA was recovered from purified preparations by phenol-chloroform extraction. Double-stranded complementary DNA was synthesized using random primers and the Super Script plasmid system for DNA synthesis and plasmid cloning (Gibco BRL). A number of clones with 600-1500 bp inserts were selected and sequenced. The sequence of this virus was compared to other viral sequences in Genbank. With about 60% of the genome sequenced, BELV was shown to have a GC content of 59.1%, and to share 66.8% sequences similarity with Oat blue dwarf virus, a member of the genus Marafivirus whose genome has been completely sequenced. A 3'-end segment of BELV had 65.6% similarity with the coat protein gene of the American isolate of Maize rayado fino Marafivirus." |
