Abstract/Contents:"We isolated and sequenced 2 genes (ViABC1 and ViABC2) from V. inaequalis, encoding drug efflux pumps belonging to the ATP-binding casette (ABC) transporter superfamily and investigated their role in fungicide resistance. ViABC1 contains 6 introns and encodes a protein of 1552 amino acids (aa). ViABC2 contains 3 introns and encodes a protein of 1490 aa. The aa sequence of the 2 ATP-binding cassettes and the 2 transmembrane domains of both genes were aligned with each other and with those from ABC transporters from other fungi. Highest sequence similarity to ViABC1 was found for AtrA from Aspergillus nidulans (64.5%). Highest sequence similarity to ViABC2 was found for AtrB from Botrytis cinerea (38.5%). Northern analysis revealed constitutive high expression of ViABC1 and ViABC2 in some myclobutanil resistant strains but not in sensitive strains, suggesting a possible role in fungicide resistance. A functional analysis of both genes is currently being investigated in yeast."
