Abstract/Contents:"In previous trials, routine PCR detection of Xanthomonas campestris pv. pelargonii (Xcp) in geraniums resulted in inconsistent sensitivity. The problem with sensitivity was partly due to the presence of putative plant-derived substances that inhibited PCR. In an effort to reduce this inhibition, various methods of extracting bacteria from plants were combined with several DNA isolation methods. Liquid nitrogen, the Meku Juice-Press, soa, and smash methods of extraction were tried. These were combined with four different DNA isolation methods: NaOH method, DNA release method (Gene Releaser, Bio Ventures), boil method, affinity binding and selective elution method (DNeasy Plant Kit, Qiagen). Of these methods, the affinity binding and selective elution method was consistently highly effective. When this method of DNA isolation was used on dilutions of sap from Xcp-infected geraniums, PCR using the primer pair, XcpM1/XcpM2 (Sulzinkski et al., 1996), was 40-fold more sensitive than ELISA."
