Abstract/Contents: | "Pseudomonas syringae pv. syringae strain B301D produces two necrosis-inducing phytotoxins, syringomycin and syringopeptin, that function as virulence determinants in the plant-pathogen interaction. These toxins are synthesized by the thiotemplate mechanism of biosynthesis involving large multi-enzymatic peptide synthetases that activate and incorporate amino acids into peptide toxins. The entire syringomycin (syr) and syringopeptin (syp) gene clusters were localized to a 135-kb DraI fragment of the B301D genome. This region includes the syp synthetase genes encoded by sypA, sypB and sypC, encompassing ~71 kb of DNA located adjacent to the ~45 kb syr cluster. In addition to the syr and syp biosynthetic genes, genes for secretion and regulation of syringomycin and syringopeptin syntesis were localized to the DraI fragment. The syr-syp cluster encompasses at least 116 kb, which represents about 2% of the P. s. pv. syringae B301D genome." |
