| Abstract/Contents: | "Dollar Spot, caused by Sclerotinia homeocarpa (S.h.) F.T. Bennett is an important disease of high maintenance turfgrass in North America. Current identification of dollar spot is based on culture morphology of S.h. on artificial media and symptom expression on host tissue. However, accurate detection may be difficult as S.h. lacks the ability to produce diagnostic reproductive propagules. The objective of this study was to develop species specific primers to provide positive identification of putative S.h. isolates. Internal transcribed spacer (ITS) 1, 5.8S and ITS 2 sequences from North American S.h. isolates were compared with isolates from the United Kingdom and closely related species. Primers for use in PCR were developed for North American isolates of S.h. based on variable ITS 1, 5.8S and ITS 2 regions between North American, British and closely related species. PCR conditions were optimized and used to identify S.h. from infected turfgrass. Species specific primers may be used in the future for the detection of dollar spot on turfgrass/thatch samples and to gain further insight into the epidemiology of dollar spot." |
