Abstract/Contents:"Tifway, an interspecific triploid hybrid [Cynodon transvaalensis Burtt-Davy x C. dactylon (L.) Pers], and its derivatives are important bermudagrass cultivars for golf courses and athletic fields in warmer temperate to tropical regions of the world. Development of a reliable Tifway tissue culture protocol is necessary to facilitate genetic transformation of these bermudagrass cultivars for further improvement. The objective of this study was to find a way to induce embryonic callus and to regenerate Tifway plants. Although callus was readily induced from Tifway vegetative tissues, they failed to regenerate into plants. Callus induction from young inflorescence was hindered by necrosis of the explant in culture medium. This problem was overcome by dipping the explants in a liquid MS medium containing 0.2% L-ascorbic acid prior to plating and by inclusion of 6 g L-1 polyvinylpolypyrrolidone (PVPP), a polphenol adsorbent, in the culture medium. The callus induction rate was 53% when segments of immature inflorescences (less than 15 mm in length) were cultured under these conditions. Two green Tifway plantlets were regenerated."
