Abstract/Contents:"Photosynthetic rate increases with ploidy level in several C3 species. The influence of ploidy level on C4 photosynthesis has not been investigated. Therefore, we are interested in the effects of polyploidy on photosynthesis and enzyme activities in C4 plants. In this study we measured rates of photosynthetic and transpiration, and activities of several key photosynthetic enzymes in leaves of naturally occurring tetraploid and octaploid Panicum virgatum (NAD-malic enzyme type) which were grown under identical conditions. The octaploid had approximately at 40% higher rate of photosynthesis/leaf area than the higher photosynthetic rates corresponded to enzyme activites that were 15 to 70% higher on a leaf area basis in the octaploid. For example, the activity of PEP carboxylase was 50% higher, RuBP carboxylase 31% higher, and NAD-malic enzyme 70% higher in the octaploid. The octaploid also had approximately 40% more chlorophyll and soluble protein than the tetraploid at all temperatures examined (20 to 40 d C). The higher photosynthetic rates corresponded to enzyme activities that were 15 to 70% higher on a leaf area basis in the octaploid. For example, the activity of PEP carboxylase was 50% higher, RuBP carboxylase 31% higher, and NAD-malic enzyme 70% higher in the octaploid. The octaploid also had approximately 40% more chlorophyll and soluble protein than the tetraploid. We are currently investigating whether this increased rate of photosynthesis in the octaploid is due to increased gene dosage."
