| Abstract/Contents: | "A double antibody enzyme-linked immunosorbent assay (ELISA) has been developed to detect Pythium Blight in turfgrass. The test is carried out in 96-well microtiter plates and utilizes a monoclonal antibody directed aginst Pythium species. The monoclonal antibody is the product of hybridoma cell line PA5IIIF11, which was produced by standard cell fusion procedures, utilizing Pythium aphanidermatum as the immunogen. The antibody binds P. aphanidermatum, P. graminicola, P. myriotylum, and P. ultimum, all of which can be involved in warm season Pythium Blight. Pythium spp. were detected in inoculated and naturally infected turfgrass samples, and the color intensity of the final reaction product of the immunoassay was directly related to the level of disease present. No cross-reactivity was observed with other common turfgrass pathogens." |
