Abstract/Contents:"Perennial ryegrass (Lolium perenne) is widely used for both turf cover and forage, whereas annual ryegrass (L. multiflorum) is only used for temporary ground cover because it does not over-winter. Unfortunately, distinguishing seeds of perennial and annual ryegrass from one another is difficult because of their similarities in weight, shape, size and colour. Previous methods to distinguish these seeds have been unreliable and timeconsuming [time-consuming]. Therefore, there is a need for an accurate and low-cost alternative method to distinguish ryegrass species. Here, a one-step protocol that combines polymerase chain reaction (PCR) and polyacrylamide gel electrophoresis (PAGE) techniques was developed to distinguish annual and perennial ryegrass seeds from one another. Based on differences between the DNA sequences in the plastid regions, a pair of primers was designed to distinguish the accessions of 26 perennial and 34 annual ryegrass using this protocol. The results clearly differentiated the two species, confirming the accuracy and sensitivity of this method."
