Abstract/Contents:"Perennial ryegrass is often associated with the fungal endophyte Epichloë festucae var. lolii. Research has shown that native endophyte associations can alter host response to abiotic and biotic stresses. However, many studies find that novel associations (non-native endophyte) can change the effect of the endophyte on plant response to stress. Most research focusing on novel inoculation technique utilizes tissue culture or inoculation into seedlings. The focus of this study was to explore inoculating the fungal endophyte into perennial ryegrass mature tiller tissue. This will allow for more rapid inoculation and allow for isogenic studies to be performed. The inoculation technique tests endophyte injection, in vitro host incubation, and differing intervals of dark. Preliminary results indicated that prolonged periods of darkness during incubation increase the likelihood of at least short-term infection."
