| Abstract/Contents: | "Recent studies indicate that photosynthesis and photorespiration can be affected by ploidy level. In most cases, this effect is due to increased levels of the bifunctional enzyme ribulosebisphosphate carboxylase/oxygenase (RuBPCase). However, isogenic diploid (2x) and tetraploid (4x) cultivars of ryegrass have been reported to possess an altered RuBPCase. A qualitative change in the enzyme at the molecular level is suggested by differences in isoelectric point and the Km and Ki for CO2 (Garrett, Nature 274:913) and in the rate of photorespiratory glycolate formation relative to photosynthesis in isolated mesophyll protoplasts and chloroplasts (Rathnam and Chollet, Plant Physiol. 65:489). Since we have been unable to confirm the reported difference in Km(CO2) between the purified, fully CO2/Mg2+-activated 2x and 4x enzymes, the kinetics of CO2/Mg2+-activation and deactivation are now under investigation. Studies are also underway to compare the physicochemical properties of the 2x and 4x RuBPCases by crystallization techniques and isoelectric focusing and electrophoresis of the native and dissociated enzymes." |
