Abstract/Contents:"The Polymerase Chain Reaction (PCR) has enabled the development of powerful genetic markers that provide ideal tools for identifying species and cultivars, and for defining genetic relationships. In this paper, the utilities of different PCR approaches are evaluated. SSRs are highly informative genetic markers that produce a single-locus DNA profile that is easily scored, but their use is limited by the need for DNA sequence knowledge. In contrast, arbitrary-primed PCR methods and AFLPs can be applied to any species without sequence knowledge, but these methods produce a multi-locus DNA profile for which scoring is more complex than for single-locus systems. PCR-based genetic markers can discriminate among closely related turfgrass cultivars, and the genetic relationships revealed by the genetic markers are congruent with other lines of evidence. While PCR-based genetic markers can demonstrate distinctiveness, they may be unsuitable for demonstrating uniformity and stability, as required for plant variety registration, particulaly for outcrossing species. The role of PCR-based genetic markers in plant variety registration and other applications remains an outstanding issue."
