Abstract/Contents:"The use of flow cytometry to rapidly estimate chromosome number or ploidy, based on nuclear DNA content, has been demonstrated for some plant species in recent years. Knowledge of the chromosome number of Cynodon plants is important, particularly in breeding improvement programs, because reported numbers range from 2n=2x=18 to 2n=6x=54. Objectives of this research were to develop flow cytometric protocol for estimating nuclear DNA content in Cynodon and to characterise its relationship to ploidy level. Nuclear DNA contents were determined for three to five clonal plants from each of four cytotypes (2n=2x=18, 2n=3x=27, 2n=4x=36, or 2n=6x=54). Flow cytometric analyses were conducted using a Becton-Dickinson FACStar^D]p^D]l^D]u^D]s flow cytometer. Nuclear suspension samples of each genotype were analysed on two dates (runs), with duplicate samples (replications) analysed on a given run. Significant (P<0.05 or 0.01) differences in DNA content existed among cytotypes and genotypes within 4x and 6x cytotypes. Mean nuclear DNA contents of 2x, 3x, 4x and 6x cytotypes were 1.11±0.04, 1.60±0.04, 2.25±0.13 and 2.80±0.14, respectively. Smaller differences were found among 4x and 6x genotypes. The results indicate flow cytometry to be a rapid and reliable means of estimating ploidy in Cynodon."
