Abstract/Contents:Studies at the University of Arkansas determined the optimum conditions for plantlet growth from calli of bermudagrass. Bermudagrass shoots were collected from fields or from sod in the greenhouse. Innermost leaves were disinfected with 70% ethanol, 1% sodium hypochlorite and autoclaved distilled water. Five explants from each leaf were planted in a petri dish with 25 ml of medium. Calli were induced on leaves from a process of adding different rates of 2,4-D and kinetin in separate conditions and mediums. Low frequency shoot formation occurred after induced calli were transferred to a Murashige-Skoog (MS) medium with .1-.5 mg/L kinetin or a hormone-free MS medium. Two plantlets were potted and grown to maturity. Calli were also induced on young influoresence to MS medium with 1 mg/L of 2,4-D and transferred to N6 medium with .25 mg/L of 2,4-D and 60g/L of sucrose. Embryo-like structures gave rise to shoots after transferring to hormone-free N6 medium. More than 11 plants were obtained from a single callus. These results show that this tissue culture system can regenerate bermudagrass callus which can produce multi-tolerant mutants, extending adaptability of bermudagrass.
