Abstract/Contents: | "Epichloe and Neotyphodium species are heritable (seed transmitted) fungal symbionts (endophytes) of cool-season grasses. Endophytes profoundly improve fitness and persistence of their hosts by providing enhanced protection from herbivory, drought, and pathogens. The lolines (saturated 1-amino-pyrrolizidines) are insecticidal alkaloids unique to grass-endophyte mutualisms. Neotyphodium uncinatum produced N-formylloline in culture, providing the first evidence that this important fitness factor is a fungal product. Furthermore, a Mendelian genetic analysis of the sexual endophyte, Epichloe festucae, indicated a single locus controlling loline alkaloid expression. Linked DNA polymorphisms (AFLP) also supported single locus control. The results of assays with insect herbivores revealed that anti-insect activity co-segregated with expression of lolines, and insect mortality correlated with loline alkaloid levels. This is the first Mendelian analysis of a fungal factor directly contributing to a plant-fungus mutualism." |
